Effects of Portulaca Oleracea Polysaccharide on Epithelial Mesenchymal Transition of Human Lens Epithelial Cells Induced by High Glucose

- Objective: To observe the effects of Portulaca oleracea polysaccharide on epithelial mesenchymal transition of immortalized human lens epithelial cells ( SRA01 / 04) induced by high glucose. Methods: SRA01 / 04 cells were cultured in vitro and randomly divided into normal control group, high glucose group and high glucose + Portulaca oleracea polysaccharide group. The cells were cultured in 0. 5 mg / ml Portulaca oleracea polysaccharide medium for 48 h, and the morphological changes of the three groups were observed under the inverted microscope at 0 h, 6 h, 12 h, 24h and 48 h. The protein expression of ZO -1, E - cadherin, α - SMA and vimentin were detected by Western blot after 48 h. Results: Compared with the control group, the cells in the high glucose group grew longer, and the number of cells decreased gradually; the morphological changes of the high glucose combined with purslane polysaccharide group and the normal group were basically the same, and there was no obvious spindle shape change with time. Compared with the control group, the expression of ZO -1, E - cadherin in high glucose group was decreased ( P < 0. 01) , the expression of α- SMA and vimentin was increased ( P < 0. 01) ; the expression of ZO -1 and E - cadherin in high glucose combined with Portulaca polysaccharide group was decreased ( P < 0. 05) , and the expression of α - SMA and vimentin was increased ( P < 0. 05) . Compared with the high glucose group, the expression of ZO -1 and E - cadherin increased in the high glucose combined with Portulaca polysaccharide group ( P < 0. 01) , while the expression of α - SMA and vimentin decreased ( P < 0. 01) . Conclusion: High glucose can induce epithelial mesenchymal transition of lens epithelial cells, and Portulaca oleracea polysaccharide can inhibit this process.
