摘要:
目的 旨在寻找术后腹腔粘连的解决办法,我们采用芍药苷抗氧化机制探索其在动物模型中降低术 后腹腔粘连形成效果。方法 选取 32 只健康雄性 SD 大鼠,随机分配为 4 组: 空白对照组、对照组、几丁糖阳性对照 组以及芍药苷组,每组 8 只。除空白对照组外其他组均构建术后腹腔粘连模型。术后第 8 天将所有动物处死。通 过肉眼评估腹腔粘连情况并评分; 检测粘连组织中纤维化 AGE 的含量、Masson's 染色; 检测 SOD - 2 及 NO 的活性, 并进一步采用 Western blotting 定量分析 SOD - 2 以及 Collenge I 的表达情况。结果 术后第8 天通过肉眼评估芍药 苷治疗后腹腔粘连形成情况,结果显示粘连评分显著轻于对照组,差异具有统计学意义( P < 0. 50) ; 芍药苷治疗后 粘连组织纤维降低,AGE、Masson's 染色、collagen 1 表达显著低于对照组( P < 0. 50) ,且粘连组织 SOD - 2 以及 NO 的表达也显著高于对照组,具有统计学差异( P < 0. 05) 。结论 芍药苷可通过促进组织抗氧化机制降低术后腹腔 粘连的形成,与临床上用于预防术后粘连形成的几丁糖比较其效果更明显,具有术后用于预防腹腔粘连潜力。
Abstract:
In order to find a solution to postoperative intraperitoneal adhesion,we use paeoniflorin an- tioxidant mechanism to explore the effect of reducing postoperative intraperitoneal adhesion in animal models. Method: Thirty - two healthy male Sprague - Dawley rats were randomly divided into 4 groups: blank control group,control group,chitosan positive control group and paeoniflorin group,with 8 rats in each group. Expect for the blank control group,postoperative intraperitoneal adhesion models were constructed in other groups and all animals were sacrificed on the 8th day after surgery. intraperitoneal adhesions were evaluated visually and scored. The content of fibrotic AGE,Mas- son's staining and the activity of SOD - 2 and NO were detected. The expression of SOD - 2 and Collenge I were further analyzed by Western blotting. Result: On day 8 postoperatively,the formation of peritoneal adhesions after paeoniflorin treatment was evaluated. The results showed that the adhesion score was significantly lower than that of the control group ( P < 0. 50) ,the difference was statistically significant ( P < 0. 50) . Paeoniflorin treatment reduced the adhesion tissue fi- bers,AGE,Masson's staining,collagen 1 expression was also significantly lower than the control group ( P < 0. 50) ,and the adhesion tissue SOD - 2 and NO expression was significantly higher than the control group,with statistical difference ( P < 0. 05) . Conclusion: Paeoniflorin can reduce the formation of postoperative intraperitoneal adhesion by promoting the antioxidant mechanism of tissues. It is more effective than the chitosan used to prevent postoperative adhesion and has the potential to prevent abdominal adhesion after operation.
