摘要:
目的 探讨临床验方前胡汤对支气管炎神经源性炎症的相关调控机制?方法 采用烟熏法建立大鼠 急性支气管炎模型,将大鼠随机分为对照组?模型组?急支糖浆组?惠菲宁组及前胡汤高?低剂量组?检测大鼠血 清 P物质(SP)?神经激肽 A(NKA)?神经激肽 B(NKB)?降钙素基因相关肽(CGRP)?神经生长因子(NGF)水平, 测定支气管肺泡灌洗液中神经源性炎症因子和肺组织中 SP和 NGF水平,肺组织中 SP受体神经激肽受体 l(NK -1)?辣椒素受体亚型 1(VR1)及 p38MAPK的 mRNA和蛋白表达水平?结果 模型组大鼠神经源性炎症物质水 平显著升高,前胡汤可明显抑制模型大鼠体内神经源性炎症物质水平,抑制作用具有一定的剂量相关性(p< 0.05)?造模可以明显增加肺组织中 NK-1及 p38MAPK的 mRNA和蛋白表达,但对于 VR1的 mRNA和蛋白表 达未见显著影响?前胡汤对 NK-1和 p38MAPK的表达量有明显抑制作用(p<0.05),对 VR1的 mRNA和蛋白 表达均未见明显影响?结论 急性支气管炎大鼠伴有神经源性炎症物质水平显著升高,前胡汤对气道神经源性 炎症具有较好的改善作用,其作用机理与调节 NGF/SP/p38MAPK信号通路有关?
Abstract:
Objective:ToexploretheregulatorymechanismofQianhuDecoctiononneurogenicinflammationofbronchitis.Methods:Theratmodelofacutebronchitiswasestablishedbysmoking.Theratswererandomlydividedintocontrolgroup,modelgroup,Jizhisyrupgroup,Huifeininggroup,QianhuDecoctionhigh-doseandlow-dosegroups.ThelevelsofsubstanceP(SP),neurokininA(NKA),neurokininB(NKB),calcitoningene-relatedpeptide(CGRP)andnervegrowthfactor(NGF)inserum,thelevelsofneurogenicinflammatoryfactorsinbronchoalveolarlavagefluid,SPandNGFinlungtissue,SPreceptorneurokininreceptorL(NK-1),capsaicinreceptorsubtype1(VR1)andp38MAPKmRNAandproteinoflungtissueweremeasured.Results:Thelevelsofneurogenicinflammatorysubstancesinthemodelgroupweresignificantlyinreased.QianhuDecoctioncouldsignificantlyinhibitthelevelsofneurogenicinflammatorysubstancesinthemodelrats,andtheinhibitioneffectshadacertaindosecorrelation(p<0.05).ThemRNAandproteinexpressionofNK-1andp38MAPKinlungtissueweresignificantlyincreased,butthemRNAandproteinexpressionofVR1werenotsignificantlyaffected.QianhuDecoctionsignificantlyinhibitedtheexpressionofNK-1andp38MAPK(p<0.05),buthadnosignificanteffectsonthemRNAandproteinexpressionofVR1.Conclusion:Thelevelsofneurogenicinflammatorysubstancesinratswithacutebronchitisaresignificantlyincreased.QianhuDecoctioncanimproveairwayneurogenicinflammation,anditsmechanismisrelatedtotheregulationofNGF/SP/p38MAPKsignalingpathway
