摘要:
目的 通过生物信息学相关技术方法探讨甲状腺功能亢进与骨质疏松之间的相互关系。方法 分别以骨质疏松及甲状腺功能亢进为关键词在相关疾病数据库(Disgenet、TTD、OMIM、Drugbank、Genecards)中筛选相关基因,去重整合后将两组预测靶基因进行映射得到关键靶点,通过STRING网站获得靶点PPI互相作用网络,根据degree值筛选出核心靶点并通过 DAVID 数据库进行GO富集分析以及 KEGG 通路分析。结果 通过相关疾病数据库搜索筛选出与骨质疏松症和甲状腺功能亢进相关靶点分别为875个和221个,映射出关键靶点99个,以degree≥35筛选出核心靶点16个并导入DAVID数据库获得44项GO富集结果(P<0.05)及45条KEGG信号通路(P<0.05)。结论 INS、IL6、ALB、TNF、IGF1等核心靶点作用于Jak-STAT通路、PI3K-Akt通路、HIF-1通路、细胞因子和细胞因子受体相互作用通路、IBD通路等多条信号通路同时对骨质疏松及甲状腺功能亢进起到调控作用,两种复杂代谢性疾病虽然各自包含众多作用靶点,但仍在分子机制上有着密切联系。应用生物信息学具有较高的置信度和参考价值,为探索疾病间关系提供了新方向与新思路。
Abstract:
Objective To explore the relationship between hyperthyroidism and osteoporosis by bioinformatics related techniques.Methods Relevant genes were screened in the related disease database (Disgenet,TTD,OMIM,Drugbank,Genecards) with osteoporosis and hyperthyroidism as key words,After de integration,the two groups of predicted target genes were mapped to obtain the key targets,the target PPI interaction network was obtained through the STRING website,the core targets were screened according to the degree value,and the go enrichment analysis and KEGG pathway analysis were carried out through the DAVID database.Results 875 and 221 targets related to osteoporosis and hyperthyroidism were screened through the search of related disease database,and 99 key targets were mapped.16 core targets were screened by degree ≥35 and imported into DAVID database.44 go enrichment results (P<0.05) and 45 KEGG signal pathways (P<0.05) were obtained.Conclusion INS,IL6,ALB,TNF,IGF1 and other core targets act on JAK STAT pathway,PI3K-Akt pathway,HIF-1 pathway,cytokine and cytokine receptor interaction pathway,IBD pathway and other signal pathways,and regulate osteoporosis and hyperthyroidism at the same time.Although the two complex metabolic diseases contain many targets,But it is still closely related to the molecular mechanism.Applied bioinformatics has high confidence and reference value,which provides a new direction and new ideas for exploring the relationship between diseases.
