摘 要:目的 观察七珠胶囊对四氯化碳(CCl4)致大鼠慢性肝损伤的保护作用,探究其作用机制。方法 SD大鼠随机分为空白对照组、模型组、阳性对照组、七珠胶囊高、中、低剂量治疗组,除空白对照组外,其余各组大鼠腹腔注射40%CCl4橄榄油溶液,2次/w,连续8 w,建立CCl4致慢性肝损伤大鼠模型,从造模成功后第一天起,连续灌胃给药,至第8w末。计算大鼠肝脏脏器指数,微板法检测血清中ALT、AST的含量,HE染色观察肝组织病理变化;生化法检测肝组织中SOD、GSH-Px活性和MDA含量; Western-blot检测肝组织TNF-α、NF-κB表达及含量变化。结果 与模型对照组比较,七珠胶囊组各剂量治疗组能显著改善肝组织病理变化,减少纤维沉积,降低血清中ALT和AST活性,并能显著升高SOD、GSH-Px活力,减少MDA含量,显著抑制肝组织中TNF-α、NF-κB表达(P<0.05)。结论 七珠胶囊对CCl4诱导的大鼠慢性肝损伤具有明显保护作用,其作用机制可能与减轻氧化应激损伤、调控TNF-α/NF-κB通路抑制症反应相关。
Abstract:
Abstract:Objective To observe the protective effect of Qizhu Capsule on chronic liver injury induced by carbon tetrachloride (CCl4) in rats and explore its mechanism.Methods SD rats were randomly divided into blank control group,model group,positive control group and high,medium and low dose treatment groups of Qizhu capsule.Except for the blank control group,rats in other groups were intraperitoneally injected with 40% CCl4 olive oil solution,twice/Week for 8 weeks.A rat model of CCl4 induced chronic liver injury was established.From the first day after successful modeling,the drug was continuously administered by gavage until the end of the 8th week.The liver organ index was calculated,the contents of ALT and AST in serum were detected by microplate method,and the pathological changes of liver were observed by HE staining;The activities of SOD,GSH-Px and the content of MDA in liver tissue were detected by biochemical method;TNF-α,NF-κB expression and content changes in liver tissue was detected by Western blot.Results Compared with the model control group,each dose of Qizhu capsule group could significantly improve the pathological changes of liver tissue,reduce fiber deposition,reduce the activities of ALT and AST in serum,significantly increase the activities of SOD and GSH-Px,reduce the content of MDA and significantly inhibit TNF-α and NF-κB expression in liver tissue (P<0.05).Conclusion Qizhu capsule has obvious protective effect on CCl4 induced chronic liver injury in rats,and its mechanism may be related to reducing oxidative stress injury and regulating TNF-α/ NF- κB pathway inhibition is related to the symptomatic response.
