摘 要:目的 观察石菖蒲挥发油吸嗅早期干预经嗅觉通路对创伤后应激障碍(Post traumatic stress disorder,PTSD)模型大鼠行为学的影响,并探索其海马GR基因相关机制。方法 采用单一延长应激造模方法建立PTSD大鼠模型,将大鼠随机分为对照组、模型组、嗅神经切断+模型组、模型+石菖蒲吸嗅组、嗅神经切断+模型+石菖蒲吸嗅组。对PTSD模型大鼠进行石菖蒲挥发油吸嗅早期干预,运用拒俘反应实验检测大鼠的易激惹程度,运用开场实验及高架十字迷宫实验检测大鼠的焦虑水平,并采用Western blot法测定大鼠海马糖皮质激素受体(Glucocorticoid receptor,GR)蛋白的表达水平。结果 经石菖蒲挥发油吸嗅早期干预,PTSD模型大鼠在开场实验中的中央活动路程百分比明显升高(P<005),在高架十字迷宫实验中的开臂进入次数百分比明显升高(P<005),在拒俘反应实验中的评分亦有所降低(P>005),表明石菖蒲挥发油吸嗅早期干预减轻了PTSD模型大鼠的易激惹程度及焦虑水平,且其作用的发挥有赖于嗅觉通路结构的完整性。另外,经石菖蒲挥发油吸嗅早期干预,PTSD模型大鼠海马GR蛋白表达下调。结论 石菖蒲挥发油吸嗅经嗅觉通路在早期干预PTSD中发挥有效作用,其作用机制可能与海马GR基因有关。
Abstract:
Abstract:Objective To observe the effect of early intervention of volatile oil from Acorus tatarinowii on behavior of PTSD model rats, and to explore the related mechanism of hippocampal GR gene. Methods PTSD rat model was established by single prolonged stress. The rats were randomly divided into control group, model group, olfactory nerve transection + model group, model + Acorus tatarinowii olfactory group, olfactory nerve transection + model + Acorus tatarinowii olfactory group. RTC was used to detect the irritability of rats, the anxiety level of rats was detected by opening experiment and elevated cross maze experiment,and the expression level of GR protein in rat hippocampus was measured by Western blot. Results The percentage of central activity distance of PTSD model rats in opening experiment was significantly increased after the early intervention of volatile oil of Acorus tatarinowii (P<0.05), the percentage of open arm entry times in elevated cross maze test increased significantly (P<0.05), and the score in rejection response test decreased (P>0.05), indicating that the early intervention of volatile oil of Acorus tatarinowii reduces the irritability and anxiety level of PTSD model rats, and its effect depends on the integrity of olfactory pathway structure.In addition, through the early intervention of volatile oil of Acorus tatarinowii, the expression of GR protein in hippocampus of PTSD model rats is down-regulated. Conclusion The olfactory pathway of volatile oil of Acorus tatarinowii plays an important role in the early intervention of PTSD. The mechanism may be related to hippocampal GR gene.
