摘 要:目的 从下丘脑内质网应激通路PERK、ATF4探讨逆灸预防大鼠亚健康状态的部分机制。方法 采用清洁级健康雄性SD大鼠,按随机数字表法分为空白组、模型组和逆灸组。逆灸组预先选用“百会”“足三里”“肾俞”穴进行逆灸(灸预处理)21 d,继而与模型组均采用束缚应激结合游泳运动(21 d)制备大鼠亚健康状态模型;空白组不做任何治疗。各组大鼠,第23 d、45 d进行旷场实验;第45 d以免疫组化法(SABC)测定下丘脑蛋白激酶样ER激酶(PERK)、转录激活因子4(ATF4)变化。结果 第23 d后,逆灸组大鼠旷场实验评分较模型组有非常明显差异(P<0.01),与空白组无明显差异(P>0.05)。第45 d后,模型组较空白组旷场实验评分减低,有非常明显差异(P<0.01);逆灸组得分高于模型组而低于空白组,均有非常明显差异(P<0.01)。第45 d后,与空白组比较,模型组、逆灸组大鼠PERK含量明显升高,模型组有非常明显差异(P<0.01),逆灸组有明显差异(P<0.05);与模型组比较,逆灸组较低,有明显差异(P<0.05)。与空白组比较,模型组、逆灸组大鼠ATF4含量也明显升高,均有明显差异(P<0.05),与模型组比较,逆灸组也较低,有明显差异(P<0.05)。结论 逆灸预防亚健康状态可能与调控下丘脑内质网应激通路PERK、ATF4相关。
Abstract:
Abstract:Objective To explore some mechanisms of inverse moxibustion in preventing sub-health in rats from the stress pathways PERK and ATF4 of hypothalamic endoplasmic reticulum.Methods Clean healthy male SD rats were randomly divided into blank group,model group and inverse moxibustion group.In the inverse moxibustion group,Baihui,Zusanli and Shenshu points were selected for inverse moxibustion (moxibustion pretreatment) for 21 days,and then the sub-health model of rats was prepared by restraint stress combined with swimming exercise (21 days);The blank group did not receive any treatment.The rats in each group were subjected to open field experiment on the 23rd and 45th days;On the 45th day,the changes of hypothalamic protein kinase like Er kinase (PERK) and transcription activator 4 (ATF4) were measured by immunohistochemistry (SABC).Results After the 23rd day,the open field test score of the reverse moxibustion group was significantly different from that of the model group (P<0.01),and there was no significant difference from that of the blank group (P>0.05).After the 45th day,the open field test score of the model group was significantly lower than that of the blank group (P<0.01);The score of inverse moxibustion group was higher than that of model group and lower than that of blank group (P<0.01).After the 45th day,compared with the blank group,the content of PERK in model group and inverse moxibustion group increased significantly,there was a very significant difference in model group (P<0.01),and there was a significant difference in inverse moxibustion group (P<0.05);Compared with the model group,the inverse moxibustion group was lower,with significant difference (P<0.05).Compared with the blank group,the content of ATF4 in model group and inverse moxibustion group was also significantly higher (P<0.05).Compared with the model group,the content of ATF4 in inverse moxibustion group was also lower (P<0.05).Conclusion The prevention of sub-health state by inverse moxibustion may be related to the regulation of hypothalamic endoplasmic reticulum stress pathway PERK and ATF4.
