摘 要:目的 探讨萆薢总皂苷(TRI)对骨关节炎(Osteoarthritis,OA)模型豚鼠的治疗效果及其可能的作用机制。方法 40只豚鼠分组,对照组(Con)、骨关节造模组(OA),萆薢总皂苷给药组(TRI)、阳性药物牛膝总皂苷组(TSA)给药组。HE染色观察各组豚鼠软骨形态;番红O-固绿染色观察各组豚鼠软骨基质的情况;免疫荧光检测各组豚鼠Ⅱ型胶原蛋白表达;酶联免疫吸附测定(ELISA)检测各组豚鼠骨关节液白细胞介素1(IL-1)、肿瘤坏死因子α(TNF-α)和基质金属蛋白酶13(MMP-13);免疫组化观察各组豚鼠骨关节尼尔样1型生长因子(Nel-like molecule 1,Nell-1)表达。结果 TRI和TSA均显著缓解OA组豚鼠骨关节炎损伤和软骨基质的退化,同时显著促进Ⅱ型胶原蛋白和Nell-1表达,降低炎性因子IL-1、TNF-α和MMP-13的表达,且TRI组治疗效果优于TSA组。结论 萆薢总皂苷可能通过促进Nell-1表达对骨关节炎发挥治疗作用。
Abstract:
Abstract:Objective To investigate the therapeutic effect of total saponins of Dioscorea bicolor (TRI) on guinea pigs with osteoarthritis (OA) and its possible mechanism. Methods 40 guinea pigs were divided into control group (Con), osteoarticular modeling group (OA), total saponins of Dioscorea bifida (TRI) group, and total saponins of Achyranthes bidentata (TSA) group. HE staining was used to observe the cartilage morphology of guinea pigs in each group; Safranine O'fixation green staining was used to observe the cartilage matrix of guinea pigs in each group; The expression of type Ⅱ collagen was detected by immunofluorescence; Enzyme linked immunosorbent assay (ELISA) was used to detect interleukin-1 (IL-1), tumor necrosis factor α (TNF-α)and matrix metalloproteinase 13 (MMP-13) in bone joint fluid of guinea pigs in each group; The expression of Neil like molecule-1 (Nell-1) in bone joints of guinea pigs in each group was observed by immunohistochemistry. Results Both TRI and TSA significantly alleviated the osteoarthritis injury and cartilage matrix degradation of guinea pigs in OA group, significantly promoted the expression of type II collagen and Nell-1, and decreased the inflammatory factors IL-1, TNF-αand MMP-13. The therapeutic effect of TRI group was better than that of TSA group.Conclusion The total saponins of Dioscorea septicerae may play a therapeutic role in osteoarthritis by promoting the expression of Nell-1.
