摘 要:目的 研究抵当芪桂汤对2型糖尿病(Type 2 diabetes mellitus,T2DM)大鼠血糖和肝组织胰岛素受体底物1(insulin receptor substrate-1,IRS-1)、糖原合成酶激酶-3β(glycogen synthase kinase3β,GSK-3β)的影响。方法 采用高糖高脂饲料联合腹腔注射小剂量的链脲佐菌素建立T2DM模型大鼠,随机分为模型组(Model组)、抵当芪桂汤高剂量组(DQG组)、抵当芪桂汤常量组(DQZ组)、二甲双胍组(Met组)、抵当芪桂汤加二甲双胍组(DQZX组)。灌胃6 w后,检测各组大鼠的空腹血糖(FBG)、糖化血红蛋白(HbA1c),同时运用HE染色观察胰腺的组织形态,PAS染色观察肝糖原合成情况,并采用免疫组化法检测T2DM大鼠肝组织中IRS-1、GSK-3β蛋白的表达。结果 药物干预6 w后,与Model相比,DQZ组、DQZX组大鼠的FBG、HbA1c均呈现不同程度降低(P<0.05或P<0.01),肝组织中IRS-1表达升高(P<0.01),GSK-3β的表达下降(P<0.05或P<0.01)。结论 抵当芪桂汤可能通过上调IRS-1表达,下调GSK-3β的水平,增强胰岛素信号传导,促进肝糖原合成,从而发挥降糖作用,二甲双胍与抵当芪桂汤联合治疗T2DM时,可增强其降糖疗效。
Abstract:
Abstract:Objective To study the effects of Didang Qigui Decoction on blood glucose,insulin receptor substrate-1 (IRS-1) and glycogen synthase kinase3β(GSK-3β)in liver tissue of type 2 diabetic rats (T2DM).Methods Type 2 diabetes model rats were established by high sugar and high fat diet combined with intraperitoneal injection of small dose of streptozotocin.They were randomly divided into model group (model group),Didang Qigui Decoction high dose group (DQG group),Didang Qigui Decoction constant group (DQG group),metformin group (Met group),Didang Qigui Decoction plus metformin group (DQZX group).After 6 weeks of intragastric administration,fasting blood glucose (FBG) and glycosylated hemoglobin (HbA1c) of rats in each group were detected.At the same time,HE staining was used to observe the tissue morphology of pancreas,PAS staining was used to observe the synthesis of liver glycogen,and immunohistochemical method was used to detect IRS-1 and GSK-3β protein expression in the liver of type 2 diabetic rats.Results After 6 weeks of drug intervention,compared with model group,FBG and HbA1c in DQZ group and DQZX group decreased in varying degrees (P<0.05 or P<0.01),the expression of IRS-1 in liver tissue increased (P<0.01),and the expression of GSK-3β was decreased (P<0.05 or P<0.01).Conclusion Didang Qigui decoction may up regulate the expression of IRS-1and down regulate GSK-3β level.It can enhance insulin signal transduction and promote liver glycogen synthesis,so as to play a hypoglycemic effect.The combination of metformin and Didang Qigui decoction can enhance its hypoglycemic effect in the treatment of T2DM.
