摘 要:目的 探讨洋金花生品、炮制品的急性毒性和亚慢性毒性。方法 急性毒性实验采用小鼠最大耐受量(MTD)灌胃法,实验第1 d,小鼠经口灌胃3次给药后,连续观察7 d,记录小鼠体重、毒性反应及死亡情况;亚慢性毒性实验采用小鼠连续灌胃给药法,每天小鼠经口灌胃给药1次,连续7 d,记录小鼠的体重、毒性反应及死亡情况。结果 洋金花生品、炮制品急性毒性实验小鼠MTD大于120 g·kg-1,最大耐受倍数为4800倍。洋金花炮制品亚慢性毒性实验20 g小鼠日给药量以体表面积法换算60 kg人日用量276.8 g,为临床用量184倍。实验中,未见中毒表现,无死亡,各组小鼠平均体重均呈上升趋势,且未有显著性差异。结论 洋金花生品、炮制品对小鼠无急性毒性,洋金花炮制品对小鼠无亚慢性毒性,洋金花炮制品在7 d内以临床用量用药具有安全性,且临床用量可以增加。
Abstract:
Abstract:Objective To investigate the acute toxicity and subchronic toxicity of the raw and processed Flos Lonicerae.Methods The acute toxicity experiment was carried out by intragastric administration of the maximum tolerance dose (MTD) in mice. On the first day of the experiment, the mice were administered orally (i.g) for three times, and then observed for seven consecutive days. The weight, toxicity and death of the mice were recorded; The subchronic toxicity test was conducted by continuous intragastric administration in mice. The mice were administered orally once a day for 7 days, and the weight, toxicity and death of the mice were recorded. Results The MTD of the acute toxicity test mice of the raw and processed flos daturae was more than 120 g·kg-1, and the maximum tolerance multiple was 4800 times. Subchronic Toxicity Test of Flos Lonicerae Preparations:The daily dose of 20g mice was 276.8g, which was 184 times of the clinical dose, converted into the daily dose of 60kg person by the body surface area method. In the experiment, there was no poisoning and no death. The average weight of mice in each group showed an upward trend, and there was no significant difference. Conclusion The crude and processed products of Flos Lonicerae have no acute toxicity to mice, and the processed products of Flos Lonicerae have no sub chronic toxicity to mice. The processed products of Flos Lonicerae are safe to use in clinical dosage within 7 days, and the clinical dosage can be increased.
