摘 要:目的 探讨调脏舒秘合剂口服给予对小鼠的急性毒性。方法 将昆明小鼠随机均分成对照组、常量组和极量组3组。灌胃给药,药物剂量按照小鼠体重20.8 g·kg-1和104 g·kg-1灌胃。于24 h内重复给药2次。观察记录14 d内小鼠体重、精神状态、活动情况、饮食量、排泄物的变化。Elisa检测肝功指标谷丙转氨酶(glutamic-pyruvic transaminase,ALT)和谷草转氨酶(glutamic-oxalacetic transaminase,AST),肾功指标尿素氮(blood urea nitrogen,BUN)和肌酐(creatinine,CRE)。HE染色法观测小鼠肝、肾、脾组织、结肠病理变化。结果 与对照组相比,常量组和极量组小鼠体重无明显变化,外观、精神状态、饮食、排泄物、一般情况均正常。解剖肉眼观小鼠肝、肾、结肠、脾,均未见明显改变,HE染色显示各组织无明显病理改变。肝功和肾功检测发现与对照组相比常量组和极量组无明显变化。结论 调脏舒秘合剂在极量104 g·kg-1的情况下未引起小鼠中毒性损伤。
Abstract:
Objective To explore the acute toxicity of Tiaozang Shubi mixture in mice after oral administration.Methods Kunming mice were randomly divided into control group,normal group and maximum group.The drug was administered by gavage at the dose of 20.8 g·kg-1 and 104 g·kg-1 respectively.Repeat administration twice within 24 hours.The changes of body weight,mental state,activity,diet and excretion of mice were observed and recorded within 14 days.Elisa was used to detect the liver function indicators of glutamic-pyruvic transaminase(ALT) and glutamic-oxalacetic transaminase(AST),and the renal function indicators of blood urea nitrogen(BUN) and creatinine(CRE).HE staining was used to observe the pathological changes of liver,kidney,spleen and colon in mice.Results Compared with the control group,the body weight of mice in the normal group and the maximum group did not change significantly,and their appearance,mental state,diet,excreta and general conditions were normal.No obvious changes were found in the liver,kidney,colon and spleen of the mice by anatomical and macroscopic observation,and no obvious pathological changes were found in the tissues by HE staining.Compared with the control group,the liver function and kidney function of the normal group and the maximum group had no significant changes.Conclusion Tiaozang Shubi Mixture did not cause toxic damage in mice at the maximum dose of 104 g·kg-1.
