摘要:
目的 治疗补肾更年方对去除卵巢大鼠血清 E 2 水平和下丘脑 EＲα 蛋白表达的影响，探讨其效应机 制。方法 将 40 只大鼠用手术去除卵巢后随机分为补肾更年方大剂量组、中剂量组、小剂量组和己烯雌酚组，并 与 10 只模型组和 10 只空白组做对照，补肾更年方大、中、小剂量组以不同剂量灌胃，模型组和空白组以生理盐 水灌胃，己烯雌酚组以己烯雌酚按 1mL/100g 的剂量灌胃，连续 45 d，结束后，取血，放免法检测血清 E 2 ，Western blot 法检测大鼠下丘脑 EＲα 蛋白表达。结果 与空白组
Abstract:
To treat the effect of Bushen Gengnian Ｒecipe on serum E2 level and expres- sion of EＲα protein in hypothalamus of ovariectomized rats and to investigate its mechanism． Method: 40 rats were randomly divided into high － dose group，middle － dose group，low － dose group and dieth- ylstilbestrol group，and were compared with 10 model groups and 10 blank groups． The high，middle and low dose groups were intragastrically administered with different doses of Bushen Gengnian Ｒecipe． The model group and the blank group were intragastrically administered with normal saline． The diethyl-stilbestrol group was administered with diethylstilbestrol at a dose of 1 mL/100 g． after 45 days，the blood was taken and the serum E2 was detected by radioimmunoassay． The expression of EＲα protein in the hypothalamus was detected by Western blot． Ｒesult: Compared with the blank group，the serum E2 level of the model group was significantly decreased，and the expression of EＲα protein in the hypothal- amus was significantly decreased (P ＜0． 01)． Compared with the model group，The high，middle and low dose groups of Bushen Gengnian Ｒecipe and diethylstilbestrol group could significantly increase the level of serum E2 in rats and increase the expression of EＲα protein in the hypothalamus (P ＜0． 05)， while the low － dose group of Bushen Gengnian Ｒecipe had no obvious effect on it (P ＞0． 05)． Conclu- sion: When improving serum E2 level in ovariectomized rats，Bushen Gengnian Ｒecipe may increase the biological effects of EＲ by affecting the expression of EＲα protein，thereby exerting therapeutic effects on menopausal syndrome．
