摘要:
目的 研究湖北海棠总黄酮对小鼠急性化学性肝损伤和急性酒精性肝损伤的保护作用。方法 将昆 明小鼠 120 只随机分组,复制两种不同肝损伤模型,每种动物模型分为 6 组:空白对照组、模型组、阳性对照组 (联苯双酯125mg·kg -1 )、湖北海棠总黄酮高、中、低剂量组(1. 68、0. 84 、0. 42g·kg -1 ),每组10 只,雌雄分开饲 养,灌胃给药。急性化学性肝损伤模型小鼠,空白对照组、模型组灌胃等体积的生理盐水,阳性组给予联苯双酯, 治疗组灌胃不同剂量湖北海棠总黄酮,连续 7 天,末次给药
Abstract:
To study the protective effect of total flavonoids of malus hupehensis on acute chemical liver injury and acute alcoholic liver injury in mice. Method: 120 Kunming mice were randomly divided into two different liver injury models. Each animal model was divided into 6 groups: blank con- trol group,model group,positive control group (biphenyldicarboxylate 125 mg·kg -1),high,medi- um and low dose groups of total flavonoids of malus hupehensis(1. 68,0. 84,0. 42g·kg -1),10 in each group,male and female were separately fed and administered by intragastric administration. As for the mice with acute chemical liver injury,the blank control group and the model group were intragastri-cally administered with the same volume of normal saline. The positive group was given bifendate. The treatment group was given different doses of total flavonoidsof malus hupehensis for 7 days and the last administration for 2 hours. Thereafter,0. 1 mL of ? 10 g -1,0. 1% CCl4 olive oil solution was intrap- eritoneally injected. As for the mice with acute alcoholic liver injury,except for the blank control group,the other groups were given 56° Beijing Erguotou (10mg·kg -1) every morning,and the total flavonoids of malus hupehensis were treated in the afternoon for 7 days. All the mice in the experimental group were taken 8 hours after the last administration,and the liver index,serum alanine aminotrans- ferase (ALT),aspartate aminotransferase (AST) content,and HE staining for liver tissue pathological changes were detected. Result: In the two models of acute liver injury,compared with the model group, different doses of total flavonoids of malus hupehensis can reduce the levels of ALT and AST in the ser- um of mice with liver injury (P <0. 01),and improve pathological damage of liver in mice. Conclusion: Total flavonoids of malus hupehensis have obvious protective effects on acute liver injury induced by CCL4 and alcohol in mice.
