摘要:
目的探讨正常体质-肾虚体质-肾虚证候动态演变过程中大鼠腮腺唾液流量?渗透压?AQP1的变化规律?方法根据中医“恐伤肾”理论,运用猫吓孕鼠方法制作仔鼠肾虚体质动物模型,在此基础上肌注氢化可的松注射液制作肾虚证候动物模型,观察正常对照组?肾虚体质组?肾虚证候组大鼠唾液流量?渗透压及腮腺AQP1的变化?结果①一般状况比较:造模期间,正常对照组大鼠体重增加明显,精神状态良好,皮毛浓密有光泽,饮食正常,大小便良好?与正常对照组比较,肾虚体质组大鼠体重略轻,精神不振,毛发光泽度欠佳,部分大鼠有脱毛现象,摄食量欠佳,大便质软,小便偏多?肾虚证候组大鼠平均体重最轻,毛发稀疏,均有毛发脱落表现,胆小易惊,喜抱团取暖,精神萎靡,活动较少,摄食量少,大便多不成形,小便量多?②唾液流量检测:与正常对照组比较,肾虚体质组唾液分泌减少,流量降低,有统计学意义(P<0.05),肾虚证候组唾液流量明显降低,具有显著的统计学意义(P<0.01);③唾液渗透压检测:与正常对照组比较,肾虚体质组及肾虚证候组唾液渗透压均较低,具有显著统计学差异(P<0.01);④腮腺组织AQP1免疫组化检测:与正常对照组比较,肾虚体质组及肾虚证候组腮腺AQP1表达均减低,具有显著统计学意义(P<0.01);与肾虚体质组比较,肾虚证候组AQP1表达降低,但无统计学差异(P>0.05);⑤腮腺组织AQP1免疫印迹检测:与正常对照组比较,肾虚体质组及肾虚证候组腮腺AQP1表达均减少,具有显著统计学意义(P<0.01);与肾虚体质组比较,肾虚证候组AQP1表达降低,有统计学差异(P<0.05)?结论伴随着“正常体质-肾虚体质-肾虚证候”的动态演变过程,大鼠唾液流量?渗透压及腮腺AQP1表达均发生了变化,而且随着肾虚状态的加重,相应指标的变化更加明显,说明了唾液的分泌确实与肾功能的盛衰密切相关,其发生的内在机制可能与腮腺AQP1表达有关?
Abstract:
Objective To investigate the change rules of parotid saliva flow, osmotic pressure, and AQP1 during the dynamic evolution among rats with normal constitution, kidney deficiency constitution and kidney deficiency syndrome. Methods According to the TCM theory of “ kidney injury fear ” , an animal model of kidney deficiency in the offspring rats was made by means of cats - scaring - pregnant mice. On this basis, an animal model of kidney deficiency syndrome was made by intramuscular injection of hydrocortisone injection. The normal control group, kidney deficiency constitution and kidney deficiency syndrome were observed in terms of their changes of salivary flow, osmotic pressure and parotid gland AQP1. Results ①General comparison: During the modeling period, the rats in the normal control group gained significant weight, good mental state, thick and shiny fur, normal diet, and good bowel movements. Compared with the normal control group, the rats in the kidney -deficiency physique group were slightly lighter, weakened, with hair gloss. Some rats had hair loss, poor food intake, soft stools and excessive urination. The rats in the kidney -deficiency syndrome group have the lightest average weight, sparse hair, all have hair loss, and they are timid and easy to be shocked. They like to hug together to keep warm with low spirt less activity, less food intake, more irregular stools, and more urination. ②Saliva flow test: Compared with the normal control group, the saliva secretion and flow of the kidney deficiency constitution group were decreased, which was statistically significant ( P <0.05) , and the saliva flow of the kidney deficiency syndrome group was significantly reduced, which was statistically significant ( P <0.01) . ) ; ③ Saliva osmotic pressure test: Compared with the normal control group, the saliva osmotic pressure of the kidney - deficiency constitution group and the kidney – deficiency syndrome group were lower with significant statistical difference ( P <0. 01) ; ④Parotid gland tissue AQP1 immunohistochemical test: Compared with the normal control group, the expression of AQP1 in the parotid gland of the kidney -deficiency constitution group and the kidney - deficiency syndrome group was reduced, which was statistically significant ( P <0.01) ; Compared with the kidney -deficiency constitution group, the expression of AQP1 in the kidney -deficiency syndrome group was reduced, but there was no statistical difference ( P >0. 05) ; ⑤ Western blot detection of parotid gland tissue AQP1: Compared with the normal control group, the expression of AQP1 in the parotid gland of the kidney deficiency constitution group and the kidney deficiency syndrome group decreased, which was statistically significant ( P <0. 01) ; Compared with the kidney deficiency constitution group, the expression of AQP1 in the kidney deficiency syndrome group decreased, with statistical difference ( P <0.05) . Conclusion Along with the dynamic evolution process of “ normal constitution, kidney deficiency constitution and kidney deficiency syndrome” , rats'saliva flow, osmotic pressure and parotid gland AQP1 expression have all changed, and with the aggravation of kidney deficiency, the changes in corresponding indicators are more obvious. It shows that the secretion of saliva is indeed closely related to the ups and downs of renal function, and its internal mechanism may be related to the expression of parotid gland AQP1.
