摘 要:目的 通过儿童社区获得性肺炎转录组分析结合网络药理学及实验验证方法,阐述麻杏石甘汤治疗儿童社区获得性肺炎的主要靶点。方法 使用R 3.5.0中limma、factoextra、dplyr、tibble、tidyr、ggplot2包进行分析及绘图,采用TCMSP、TCMID、PharmMapper数据库获取麻杏石甘汤成分及靶点。使用Elisa进行IL-6和TNF-α检测及Western Blot检测AK1、CYP1B1和MGAM。结果 社区获得性肺炎组与对照组共有335个差异表达基因;麻杏石甘汤可减少A549细胞的炎症损伤;麻杏石甘汤可增加AK1的水平,减少CYP1B1和MGAM的水平,从而起到调控社区获得性肺炎的作用。结论 麻杏石甘汤可调节社区获得性肺炎相关靶点及炎症水平。
Abstract:
Abstract:Objective To elucidate the main target of Maxing Shigan decoction in the treatment of children’s community acquired pneumonia through Transcriptome analysis combined with network pharmacology and experimental verification methods.Methods R 3.5.0 were analyzed and plotted using limma,factoextra,dplyr,tible,tidyr,and ggplot2 packages.The components and targets of Maxing Shigan decoction were obtained using TCMSP,TCMID,and PharmaMapper databases.Using Elisa for IL-6 and TNF-α Detection and Western Blot detection of AK1,CYP1B1,and MGAM.Results There were 335 differentially expressed genes between the community acquired pneumonia group and the control group;Maxing Shigan decoction can reduce the inflammatory damage of A549 cells;Maxing Shigan decoction can increase the level of AK1,reduce the levels of CYP1B1 and MGAM,and thus play a role in regulating community acquired pneumonia.Conclusion Maxing Shigan decoction can regulate the targets and inflammation levels related to community acquired pneumonia.
