摘要:
目的 观察复方蜥蜴散不同微粒组合剂对胃癌前病变( PLGC) 模型大鼠血管内皮生长因子( VEGF) 和缺氧诱导因子( HIF - 1α) 表达的影响,探讨该方干预 PLGC 的机制。方法 SPF 级、4 - 6w 龄雄性 SD 大鼠 120 只,随机分为维酶素对照组 A、PLGC 模型对照组 B、复方蜥蜴散不同微粒组合剂高剂量组 C、中剂量组 D、低剂量 组 E、空白对照组 F,20 只/组。除 F 组外,其余各组均以 MNNG,综合饥饱失常、乙醇等灌服 24w 后病检以证实 PLGC 大鼠模型制备成功。各治疗组用药 12w 后观察大鼠胃黏膜病理改变情况; 免疫组化测胃体 HIF - 1α 和 VEGF 水平。结果 VEGF 和 HIF - 1α 水平在各模型组与 F 组比较,P < 0. 05,AOD 值降低; 各治疗组与 B 组比 较,P < 0. 05,AOD 值降低明显; C、D、E 组与 B 组比较,P < 0. 05,以 C 组 AOD 值最小。结论 复方蜥蜴散可改善 PLGC 模型大鼠胃黏膜病理,阻断大鼠癌变趋势,其机制可能与该方影响 HIF - 1α 和 VEGF 表达有关。
Abstract:
Objective:To observe the effect of different combined reagents of compound lizard powder on the expression of vascular endothelial growth factor ( VEGF) and hypoxia - inducible factor ( HIF - 1α) in gastric precancerous lesions ( PLGC) rats and to explore the mechanism of the intervention. Method: A total of 120 SPF and 4 - 6w male SD rats were randomly divided into six groups: Vatacoenayme A control group A,PLGC model control group B,different combined reagents of compound lizard powder,high dose group C,middle dose group D,low dose group E,blank control group F,with 20 cases in each. Except for group F,the rest of all the groups confirmed successful preparation of PLGCrat models 24w after MNNG,comprehensive hunger disorders,administration of ethanol and other tests. The pathological changes of gastric mucosa in the rats were observed after 12W,and the levels of HIF - 1 alpha and VEGF in the gastric tissue were measured by immunohistochemistry. Result: The levels of VEGF and HIF - 1α in each model group were sig- nificantly lower than those in group F ( P < 0. 05) ,AOD decreased significantly,and AOD decreased significantly in each treatment group compared with those in group B,p < 0. 05,while Group C,D and E were compared with group B,P < 0. 05,AOD was the smallest in Group C. Conclusion: Compound lizard powder can improve gastric mucosal pathology in rats with PLGC and block the carcinogenesis in them. The mechanism may be related to the influence of HIF - 1α and VEGF expression.
