摘 要:目的 探讨王庆国教授经验方复方参柏煎剂对慢性湿疹ICR小鼠的治疗效果及其作用机制。方法 将小鼠随机分为6组:空白组、模型组、阳性药对照组、复方参柏煎剂低剂量组、中剂量组、高剂量组,每组7只。模型组采用2,4-二硝基氯苯(DNCB)致敏建立小鼠耳朵慢性湿疹模型;复方参柏煎剂低、中、高剂量组分别给予低、中、高剂量的复方参柏煎剂(浓度分别为0.16 g·mL-1、0.32 g·mL-1、0.64 g·mL-1)干预,阳性药组给予复方醋酸地塞米松乳膏干预。观察各个组别小鼠的耳重量及厚度,并借助HE法观察其病理变化,ELISA法检测各组血清及组织中细胞因子的含量。结果 与模型组比较,复方参柏煎剂高、中、低剂量组均能降低慢性湿疹模型小鼠的耳肿胀度,降低小鼠慢性湿疹模型血清内IL-4的含量,升高IFN-γ的含量;降低小鼠慢性湿疹模型局部组织中IL-4和IL-17的含量。结论 复方参柏煎剂对小鼠慢性湿疹具有明显的治疗作用,主要通过降低炎症反应和调整Th1/Th2的比例失衡,也就是通过直接抗炎作用和间接免疫调节作用达到治疗慢性湿疹的效果。
Abstract:
Abstract:Objective To investigate the therapeutic effect and mechanism of Professor WANG Qingguo's empirical formula compound Shenbai decoction on ICR mice with chronic eczema.Methods Mice were randomly divided into 6 groups:blank group,model group,positive drug control group,low dose group,medium dose group and high dose group,with 7 mice in each group.The model group was sensitized with 2,4-dinitrochlorobenzene (DNCB) to establish the mouse ear chronic eczema model;The low,medium and high dose groups of compound Shenbai decoction were intervened with low,medium and high dose of compound Shenbai decoction (the concentrations were 0.16 g·mL-1,0.32 g·mL-1 and 0.64 g·mL-1 respectively),and the positive drug group was intervened with compound dexamethasone acetate cream.The ear weight and thickness of each group of mice were observed,and the pathological changes were observed by HE method.The content of cytokines in serum and tissue was detected by ELISA.Results Compared with the model group,the high,medium and low dose groups of compound Shenbai decoction can reduce the degree of ear swelling of chronic eczema model mice,reduce the content of IL-4 and increase IFN-γ in serum of chronic eczema model mice;Reduce the contents of IL-4 and IL-17 in local tissues of mouse chronic eczema model.Conclusion Compound Shenbai decoction has obvious therapeutic effect on chronic eczema in mice,mainly by reducing inflammatory reaction and adjusting the imbalance of Th1/Th2 ratio,that is,through direct anti-inflammatory effect and indirect immune regulation.
